DNAやRNAといった核酸の検出は、生命科学研究から臨床診断、バイオテロ対策に至るさまざまな分野において極めて重要である。DNAは、遺伝情報の運び手としての役割を果たすだけでなく、数十年間からは相補配列のハイブリダイゼーションのおかげで、優れた認識ツールとしても登場するようになった。つまり、プローブとして相補配列を用いることによって、DNA鎖を効率よく検出できるのである。この検出法は、溶液中ではうまくいくが、表面にプローブを並べる方式では難しくなる。今回、上海応用物理研究所(中国)のChunhai Fanらは、ナノ構造骨格を持つプローブを用いたプローブ―標的認識プロセスにおける最近の進展について概説している。検出用のプローブ構造は、直線状一本鎖DNAから一本鎖ループ構造、そしてさらに複雑な骨格構造(四面体など)へと進化してきた。骨格構造を利用すると、隣接するDNAプローブ同士が絡まりにくくなる一方で、プローブの配置や接近性を精密に制御できるようになる。

The detection of nucleic acid – DNA and RNA – is crucial in various fields from life sciences research to clinical diagnosis and anti-bioterrorism. Beside its role as a carrier of genetic information, over the past few decades DNA has also emerged as an excellent tool for recognition, owing to the hybridization of complementary sequences. Thus, an efficient way to detect a DNA strand is to use its complementary sequence as a probe. Although this works well in solution, it becomes more of a challenge when the probes are arranged on a surface. Chunhai Fan and co-workers at the Shanghai Institute of Applied Physics review recent developments in probe-target recognition processes involving nanostructured-scaffold probes. Such structures have evolved from a single, linear strand of DNA to a single-stranded loop conformation, and into more complex scaffolds such as tetrahedra. The latter offers precise control over the positioning and accessibility of the DNA probes, while preventing adjacent probes from entangling.
